**19.28z.HN8 CAR-T** is an engineered CAR-T cell therapy featuring an anti-CD19 CAR construct with a CD28ζ signaling domain, enhanced by overexpression of humanin-like 8 (HN8). HN8 overexpression improves CAR-T cell function, reduces exhaustion, and promotes a less exhausted, more functional phenotype, addressing limitations in standard CD28-based CAR-T therapies like increased cytokine release syndrome (CRS). The base 19-28z CAR targets CD19 on B-cell malignancies, enabling T-cell mediated tumor destruction, with design optimizations such as CD8α hinge for reduced toxicity while maintaining efficacy. Developed using lentiviral transduction of patient-derived αβ T cells, it shows promising expansion and transduction efficiency in preclinical settings.
