1907B is a novel, ultralong-acting dual agonist of the glucagon receptor (GCGR) and glucagon-like peptide 1 receptor (GLP1R). Developed by researchers at Sun Yat-Sen University, it is being investigated for its potential to treat intestinal fibrosis, a common complication of Crohn's Disease. The drug's mechanism involves the metabolic regulation of epithelial cells; specifically, it normalizes the ATP production rate of glycolysis and reduces levels of histone H3K9 lactylation (H3K9la) on the promoters of fibrotic genes. By activating both GCGR and GLP1R signaling, 1907B has demonstrated the ability to ameliorate epithelial-mesenchymal transition (EMT) and intestinal fibrosis in preclinical models of chronic colitis.
