1928z-GLUT1 CAR T cells are an experimental cellular immunotherapy developed at Memorial Sloan Kettering Cancer Center. These cells consist of T cells engineered to express a second-generation chimeric antigen receptor (CAR) targeting CD19, featuring CD28 costimulatory and CD3ζ signaling domains (19-28z), further modified to constitutively overexpress the glucose transporter GLUT1 (SLC2A1). This metabolic engineering strategy aims to overcome the nutrient-depleted tumor microenvironment, where competition for glucose often impairs CAR T cell function. By increasing glucose uptake, GLUT1 overexpression enhances both glycolysis and oxidative phosphorylation, promoting superior T cell expansion, metabolic fitness, and therapeutic efficacy compared to standard 19-28z CAR T cells, particularly in glucose-limiting conditions.
