Overview

1928z-LNGFR CAR T cells are a CD19-targeted chimeric antigen receptor (CAR) T cell therapy primarily utilized as a benchmark or control construct in preclinical and clinical research. Developed at Memorial Sloan Kettering Cancer Center, these cells are genetically engineered to express a second-generation CAR consisting of a CD19-binding domain, a CD28 costimulatory domain, and a CD3-zeta signaling domain (the '19-28z' design). Additionally, they express a truncated low-affinity nerve growth factor receptor (LNGFR) which serves as a non-immunogenic surface marker for cell selection and tracking. This construct has been extensively used to evaluate the efficacy and metabolic fitness of novel CAR designs, such as those incorporating metabolic enhancements like GLUT1 overexpression, in treating B-cell malignancies including acute lymphoblastic leukemia (ALL) and lymphomas.
