192IgG-saporin is a chemically conjugated **antibody-drug conjugate** consisting of the mouse monoclonal antibody 192-IgG and the ribosome-inactivating protein **saporin**. The 192-IgG binds specifically to the rat low-affinity neurotrophin receptor (p75), richly expressed on cholinergic neurons in the basal forebrain. When administered intracerebroventricularly or locally in rat brain, this immunotoxin is selectively internalized by p75-expressing cholinergic neurons, resulting in highly specific and permanent ablation of these neurons through the ribosome-inactivating action of saporin. This selective neurotoxin is commonly used as a research tool to model **Alzheimer's disease–associated cholinergic deficits**, study basal forebrain cholinergic system functions, and evaluate mechanisms of neurodegeneration and neuroprotection. Its action leads to depletion of cholinergic neurons in the medial septum, diagonal band of Broca, and nucleus basalis of Meynert, impairing memory and cognition in animal models[1][3][5][6][9].
