Overview

1alpha-Hydroxyvitamin D5 (CARD-024) is a synthetic analog of vitamin D, specifically a less toxic VDR (Vitamin D receptor) agonist designed to suppress proliferation and induce differentiation and apoptosis in certain cancer cells, particularly breast cancer cells that express the VDR. It has been investigated primarily as an investigational small molecule for its potential chemopreventive and therapeutic effects in oncology, with preclinical studies showing growth inhibition of VDR-positive human breast cancer cells both in vitro and in vivo. The compound upregulates VDR expression at the mRNA and protein levels, leading to modulation of gene expression via vitamin D response elements. Its maximum clinical trial phase reached is Phase I, where it was studied for safety and pharmacokinetics[1][2][3][5][6].
