1B7 is a fully human monoclonal antibody with dual specificity for the gangliosides GD2 and GM2. It was isolated from patients immunized with a GD2 lactone-keyhole limpet hemocyanin (GD2L-KLH) conjugate vaccine. Developed by MabVax Therapeutics in collaboration with Memorial Sloan Kettering Cancer Center, 1B7 exhibits high affinity for GD2 (~1 nM) and moderate affinity for GM2 (~370 nM). The antibody mediates anti-tumor activity through complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies in xenograft models have demonstrated its therapeutic potential against neuroblastoma, sarcoma, and melanoma, where it significantly inhibited tumor growth and improved survival rates.
