The 1c-SApNP HIV-1 vaccine is a protein subunit vaccine candidate that utilizes a one-component self-assembling protein nanoparticle (1c-SApNP) platform. Developed primarily by researchers at Scripps Research, this technology involves the genetic fusion of a stabilized HIV-1 envelope (Env) trimer (such as BG505 SOSIP) to a nanoparticle-forming protein subunit (e.g., I3-01 or ferritin). This fusion allows the vaccine to self-assemble into a highly ordered, multivalent nanoparticle that displays multiple copies of the Env trimer on its surface. The primary mechanism of action is to mimic the repetitive structural patterns of natural viruses, which enhances the cross-linking of B-cell receptors and promotes the induction of broadly neutralizing antibodies (bNAbs). The one-component design is intended to improve stability, simplify manufacturing, and ensure high antigenic integrity compared to more complex two-component systems.
