1C10 is a murine monoclonal antibody agonist targeting CD40, a costimulatory protein found on antigen-presenting cells such as dendritic cells. It is widely used in preclinical oncology research to stimulate antitumor immune responses. The mechanism of action for 1C10 requires the engagement of the inhibitory Fc gamma receptor IIb (FcγRIIb), which acts as a scaffold to facilitate the cross-linking and clustering of CD40 receptors necessary for downstream signaling. In models of non-muscle-invasive bladder cancer (NMIBC), intravesical administration of 1C10 has demonstrated the ability to reduce tumor burden and induce systemic, long-term antitumor immunity mediated by CD8+ T cells and conventional type 1 dendritic cells (cDC1). This murine antibody served as the foundational model for developing Fc-optimized human CD40 agonists like 2141-V11.
