Drug intelligence / Profile preview
1D-142 is a novel, small-molecule guanidine-based inhibitor of the Rho family GTPase RAC1. It acts as a protein-protein interaction inhibitor that blocks the interaction between RAC1 and its guanine nucleotide exchange factor (GEF) Tiam1 (T-cell lymphoma invasion and metastasis 1), thereby preventing RAC1 activation. Originally developed for oncology indications, including non-small cell lung cancer (NSCLC) and hepatocellular carcinoma (HCC), 1D-142 has been shown to inhibit tumor cell proliferation, migration, and metastasis, and to induce apoptosis. More recently, research has demonstrated its therapeutic potential in acute liver failure (ALF) and hepatitis, where it mitigates oxidative stress, reduces reactive oxygen species (ROS) production in hepatocytes, decreases inflammatory cytokine release, and limits immune cell infiltration.
