1D11.16 (often referred to simply as 1D11) is a murine IgG1 monoclonal antibody that acts as a pan-specific neutralizer of all three mammalian isoforms of transforming growth factor-beta (TGF-β1, TGF-β2, and TGF-β3). Originally developed by Genzyme, 1D11.16 is widely utilized as a preclinical research tool and therapeutic candidate in animal models of cancer, fibrosis, and inflammation. By binding and neutralizing active TGF-β, the antibody blocks downstream Smad signaling, thereby reversing TGF-β-mediated immunosuppression, inhibiting fibrogenesis, and enhancing the efficacy of radiotherapy and cancer vaccines. It served as the parent antibody for the development of the humanized clinical candidate fresolimumab (GC1008).
