Overview

1D3HMCys TCR-modified T cells are autologous peripheral blood T cells engineered to express a murinized, gene-optimized T-cell receptor (TCR) targeting the HLA-A*02:01-restricted MART-1(26-35) epitope, a melanoma antigen recognized by T cells. The 1D3HMCys TCR contains murine constant regions to improve expression and reduce mispairing, and incorporates an extra cysteine for enhanced stability, plus a P2A peptide linker for equal expression of both chains. These T cells are expanded ex vivo with IL-7 and IL-15 to generate products with a less-differentiated, memory stem/central memory phenotype. Developed as a form of adoptive cell therapy for metastatic melanoma, the therapy demonstrated in clinical trials both feasibility and potent antitumor activity, but was associated with dose-dependent, on-target off-tumor toxicities, including dermatitis, uveitis, hearing loss, and cytokine release syndrome, leading to early termination of trials[1][2][3].
