This is a combination therapy consisting of three components: - 1E10-Alum (also known as Racotumomab-alum), an anti-idiotype vaccine designed to induce an immune response against N-glycolyl GM3 ganglioside, a tumor-associated antigen found in various cancers. - Cyclophosphamide, an alkylating agent that interferes with DNA replication and cell division, commonly used as chemotherapy. - Methotrexate, an antimetabolite and antifolate agent that inhibits dihydrofolate reductase, disrupting DNA synthesis and cell proliferation. The combination is experimental and has been investigated primarily for its potential synergistic effects in cancer treatment. The rationale is to combine the immunostimulatory effect of the vaccine (1E10-Alum) with the cytotoxic/antiangiogenic properties of low-dose metronomic cyclophosphamide and methotrexate. Metronomic chemotherapy refers to frequent administration of chemotherapeutic agents at low doses without extended breaks, aiming for antiangiogenic effects while minimizing toxicity[1][2][3][4]. This approach has shown promise in metastatic breast cancer settings.
