1F5 is a monoclonal antibody (mAb) developed as a potential therapeutic against Nipah virus (NiV) and Hendra virus (HeV), both highly pathogenic henipaviruses. It functions by targeting the fusion (F) glycoprotein present on the surface of these viruses. By binding to a conserved quaternary epitope on the F glycoprotein, 1F5 effectively locks the protein in its prefusion conformation, thereby inhibiting membrane fusion and preventing viral entry into host cells. Preclinical studies have demonstrated its potent neutralizing activity and a 100% success rate in providing protection, even when administered several days post-infection. Humanized versions of 1F5 have been developed, and it has shown to be more effective than the previously identified mAb m102.4, which targets the G glycoprotein. Its development is supported by the U.S. DoD Joint Program Executive Office for Chemical, Biological, Radiological and Nuclear Defense and CEPI.
