**1R-2b** is an experimental, tumor-targeted immunocytokine developed by Immunomedics using its Dock-and-Lock platform. It is a single covalent antibody-cytokine complex consisting of the humanized anti-insulin-like growth factor 1 receptor antibody hR1 linked to four interferon alfa-2b molecules. The hR1 component binds and down-regulates insulin-like growth factor 1 receptor, while the interferon alfa-2b payload activates type I interferon signaling. In renal cell carcinoma cell models, 1R-2b inhibited tumor-cell growth at picomolar concentrations and showed synergistic in-vitro activity with the mTOR inhibitor temsirolimus. It has been publicly reported as a preclinical candidate for renal cell carcinoma.
