Overview

1RG-CART is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by Cancer Research UK for the treatment of relapsed or refractory neuroblastoma. The therapy consists of patient-derived T-cells transduced with a gamma-retroviral vector encoding a humanized anti-GD2 single-chain variable fragment (scFv) derived from the K666 antibody, coupled with CD28 and CD3ζ signaling domains. Additionally, the construct includes the RQR8 suicide gene, which allows for the selective depletion of the CAR T-cells using rituximab if severe toxicity occurs. The primary target is disialoganglioside (GD2), which is highly and homogeneously expressed on neuroblastoma cells. The therapy is currently being evaluated in a Phase I clinical trial (NCT02761915) to assess safety and feasibility.
