Overview

1S-1-acetoxychavicol acetate (ACA) is a naturally derived small molecule phenylpropanoid isolated from the rhizomes of Alpinia conchigera Griff. and Alpinia galanga (family Zingiberaceae). It is a research compound primarily investigated for its anticancer, antiviral, and anti-inflammatory properties. ACA's mechanism of action involves the inhibition of exportin-1 (XPO1/CRM1), which blocks the nuclear export of proteins and viral RNAs, and the inhibition of NADPH oxidase (Nox) and proteasomal chymotrypsin-like activity. These actions lead to the suppression of the NF-κB signaling pathway, induction of apoptosis, and cell cycle arrest at the G0/G1 phase. Preclinical studies have demonstrated its efficacy against various human tumor cell lines, including oral squamous cell carcinoma, non-small cell lung cancer (NSCLC), and breast cancer, as well as its potential in treating androgenetic alopecia and viral infections like HIV-1 and influenza.
