A combination prebiotic supplement comprising **2′-fucosyllactose (2′-FL)**, a prevalent human milk oligosaccharide (HMO), and **resistant starch (RS)**, a fermentable dietary fiber. The combination is designed to target the gut microbiota, specifically to enhance the production of short-chain fatty acids (SCFAs) such as acetate and butyrate in the distal colon. By increasing systemic SCFA availability, the supplement aims to modulate host energy and substrate metabolism, potentially improving insulin sensitivity and lipid profiles. Clinical research conducted by Wageningen University and Maastricht University (NCT04795804) has demonstrated that subacute supplementation can increase systemic SCFA concentrations and lower plasma free fatty acids (FFAs) in lean individuals, although metabolic benefits appear to be phenotype-specific and were less pronounced in individuals with prediabetes and obesity.
