Overview

2,5-dichloro-N-(3,5-dibromo-4-hydroxyphenyl)benzamide (DrugBank DB07694) is an experimental small molecule that acts as a potent kinetic stabilizer of the protein transthyretin (TTR). The compound functions by binding to the thyroxine (T4) binding sites at the dimer-dimer interface of the TTR tetramer, effectively increasing the activation energy barrier for tetramer dissociation. Since the dissociation of the native tetramer into amyloidogenic monomers is the rate-limiting step in transthyretin amyloidosis (ATTR), this stabilization prevents the subsequent misfolding and aggregation of TTR into amyloid fibrils. While it has been extensively used as a research tool to study the structural biology of TTR and the mechanism of amyloidogenesis inhibition, it has not progressed into clinical development by a pharmaceutical sponsor.
