Overview

**2-methyl-3-butenyl-1-pyrophosphate (2M3B1-PP)** is a synthetic pyrophosphomonoester antigen developed to selectively and potently stimulate human Vγ9Vδ2 T cells[1][2][3][4][5]. This molecule mimics naturally occurring isoprenoid pyrophosphate intermediates and is recognized by γδ T cell receptors (TCRs), especially Vγ9Vδ2, which are known to mediate tumor cytolysis and are a target for cancer immunotherapy[1][3][4]. **Mechanism of action:** 2M3B1-PP stimulates Vγ9Vδ2 T cells by acting as a non-peptide phosphoantigen, triggering TCR-dependent activation, proliferation, and resulting in strong cytotoxic activity against tumor cells[3][4][5]. The mechanism involves direct binding to the TCR and involves the interaction with BTN3A1, a butyrophilin protein, facilitating conformational change sensed by the Vγ9Vδ2 TCR[1]. The drug is used as an immunostimulant ex vivo and in vitro to expand and activate γδ T cells, which are then deployed in adoptive cell therapy protocols for various cancers, including hepatocellular carcinoma, ovarian cancer, breast cancer, and renal cell carcinoma[1][2][3][4].
