Overview

2-methyldihydrexidine is a selective, high-affinity full agonist at the dopamine D1 and D5 receptors. It is a derivative of dihydrexidine, which was the first non-catechol high-affinity full D1 agonist. Developed primarily as a research tool to investigate the dopaminergic system's role in cognitive functions, it has been studied for its potential in treating symptoms of attention-deficit/hyperactivity disorder (ADHD) and other cognitive impairments. Research indicates that 2-methyldihydrexidine can improve temporal order memory and spatial working memory in rodent models, particularly when co-administered with α2A-adrenergic agonists like guanfacine, suggesting a synergistic relationship between D1 and α2A receptor activation in the prefrontal cortex.
