Overview

21.15.GPC3-CAR T cells, also known as CO-EXIST T cells, are an autologous CAR-T cell therapy genetically engineered to target Glypican-3 (GPC3), a cell-surface protein highly expressed in various solid tumors. These cells are "armored" through the co-expression of interleukin-15 (IL-15) and interleukin-21 (IL-21) to enhance their expansion, persistence, and anti-tumor activity within the immunosuppressive tumor microenvironment. Additionally, the therapy incorporates an inducible caspase 9 (iCasp9) safety switch, which allows for the rapid elimination of the infused T cells upon administration of the small-molecule dimerizer AP1903 (rimiducid) in the event of severe adverse effects. This investigational product is being developed for the treatment of GPC3-positive solid tumors, including hepatocellular carcinoma and several pediatric cancers.
