Drug intelligence / Profile preview
211At-anti-CD44v6 VHH dimer is a preclinical-stage targeted alpha-particle radiopharmaceutical being developed by West China Hospital of Sichuan University. It consists of a CD44v6-specific single-domain antibody (VHH) dimer conjugated with the alpha-emitting radionuclide astatine-211 (211At). The therapeutic is designed for local intrapleural administration to treat malignant pleural effusion (MPE) in lung adenocarcinoma (LUAD). It specifically targets and depletes SPP1+/CD44v6+ tumor-associated macrophages (TAMs) in the immunosuppressive tumor microenvironment. By selectively inducing DNA double-strand breaks and immunogenic cell death (ICD) in these TAMs, the agent triggers cGAS-STING pathway activation and subsequent CD8+ T-cell-mediated antitumor immunity.
