212-Pb-RMV-GPC3-VH is a preclinical-stage targeted alpha therapy (TAT) being co-developed by RadioMedix and Vect-Horus for the treatment of pancreatic ductal adenocarcinoma. The drug consists of a Lead-212 (Pb-212) radiolabeled VHH nanobody (single-domain antibody fragment) directed against Glypican-3 (GPC3), a cell surface protein overexpressed in pancreatic tumors. The mechanism of action involves the nanobody binding to GPC3-expressing cells and delivering high-energy alpha radiation directly to the tumor site, causing lethal double-strand DNA breaks. This radioligand therapy approach leverages the small size and high specificity of nanobodies for rapid tumor penetration and clearance, combined with the potent, short-range cell-killing ability of Pb-212 alpha particles.
