Overview

**212Pb-376.96** is a radioimmunotherapy agent consisting of the monoclonal antibody 376.96 conjugated to **lead-212 (212Pb)**, an alpha-emitting radionuclide. The antibody specifically targets **B7-H3 (CD276)**, a cell-surface protein overexpressed in several malignancies, notably **pancreatic ductal adenocarcinoma (PDAC)** and ovarian cancer. By delivering cytotoxic alpha particles directly to tumor cells, 212Pb-376.96 causes double-stranded DNA breaks, leading to cell death. Preclinical studies have demonstrated specific binding and high cytotoxicity to PDAC cells and cancer-initiating cells, markedly inhibiting tumor growth in xenograft mouse models. 212Pb-376.96 displays greater efficacy compared to isotype-matched controls and holds promise both as monotherapy and in combination with chemotherapy in PDAC models[1][5][7].
