Drug intelligence / Profile preview
212Pb-anti-LRRC15 antibody is a preclinical targeted alpha therapy (TAT) radiopharmaceutical consisting of a monoclonal antibody targeting leucine-rich repeat-containing protein 15 (LRRC15) labeled with the alpha-emitting radioisotope Lead-212 (212Pb). LRRC15 is a transforming growth factor-beta (TGF-β)-driven cell surface protein highly expressed on cancer-associated fibroblasts (CAFs) within the tumor microenvironment and on certain mesenchymal-derived cancer cells, while showing minimal expression in normal tissues. The targeting antibody (such as DUNP19 / LNTH-2403) binds to LRRC15 and is rapidly internalized. Lead-212 acts as an in vivo alpha-particle generator, decaying to produce alpha-emitting daughters (such as Bismuth-212) that deliver highly cytotoxic, localized radiation to the tumor and its supportive stroma, sparing surrounding healthy tissue. Developed by researchers at Washington University in St. Louis, this radioconjugate is being evaluated in preclinical models of colon cancer and other solid tumors.
