212Pb-CEA PRIT is an advanced alpha radioimmunotherapy platform being developed through a collaboration between Orano Med and Roche. It utilizes a pre-targeted radioimmunotherapy (PRIT) approach to treat solid tumors overexpressing Carcinoembryonic Antigen (CEA), such as colorectal, pancreatic, gastric, and non-small cell lung cancers. The treatment involves a two-step regimen: first, a tumor-targeting bispecific antibody is administered to accumulate on CEA-positive tumor cells; second, a radioligand carrying the Lead-212 (212Pb) radioactive payload is administered. The radioligand binds to the pre-localized bispecific antibody, delivering high-energy alpha radiation directly to the tumor while minimizing exposure to healthy tissues. This clearing agent-independent regimen is designed to optimize the therapeutic window, particularly given the 10.6-hour half-life of 212Pb.
