212Pb-DARPin is a targeted alpha therapy (TAT) platform and discovery-stage program developed through a collaboration between Orano Med and Molecular Partners. The technology combines Designed Ankyrin Repeat Proteins (DARPins)—small, robust proteins engineered for high-affinity binding to tumor antigens—with lead-212 (212Pb), a rare alpha-emitting radioisotope. Upon binding to its specific molecular target on cancer cells, the 212Pb payload undergoes decay, releasing high-energy alpha particles that cause irreparable double-strand DNA breaks within a short range (50-100 µm). This mechanism allows for potent cell killing while sparing adjacent healthy tissue. The platform is designed to address various solid tumors by swapping the DARPin targeting moiety for different cancer-associated proteins. This specific program represents discovery-stage assets where the molecular targets have not yet been publicly disclosed.
