212Pb-DARPin-DLL3 (MP0712) is a preclinical-stage targeted alpha therapy (TAT) being developed by Orano Med in partnership with Molecular Partners. The drug consists of a Designed Ankyrin Repeat Protein (DARPin) binder specific for Delta-like ligand 3 (DLL3) conjugated to a Lead-212 (212Pb) radioactive payload. DLL3 is an inhibitory Notch ligand that is highly expressed on the surface of small-cell lung cancer (SCLC) and other neuroendocrine tumor cells, while remaining largely absent in healthy tissues. By utilizing the DARPin platform, the therapeutic achieves high specificity and rapid tumor penetration. The 212Pb isotope acts as an alpha-particle emitter, delivering high-energy radiation to induce double-strand DNA breaks in tumor cells, leading to cell death with minimal impact on surrounding healthy tissue.
