Overview

212Pb-LNC1011M is a targeted alpha-therapy (TAT) radiopharmaceutical developed by LNC Pharma for the treatment of prostate cancer. The drug candidate consists of a small-molecule ligand, LNC1011M, designed to bind with high affinity to the prostate-specific membrane antigen (PSMA), which is frequently overexpressed in prostate cancer cells. This ligand is conjugated to Lead-212 (212Pb), a radioisotope that serves as a source of alpha particles. Upon binding to PSMA and subsequent internalization into the tumor cell, the decay of Lead-212 releases high-energy alpha radiation. These alpha particles cause lethal double-strand DNA breaks within a very short range, effectively killing the target cancer cells while sparing surrounding healthy tissue. 212Pb-LNC1011M is currently in preclinical development.
