212Pb-PS0001 is a targeted alpha therapy (TAT) radiopharmaceutical developed by AdvanCell for the treatment of prostate cancer. It consists of the alpha-emitting radioisotope lead-212 (212Pb) conjugated to a high-affinity ligand that specifically targets the prostate-specific membrane antigen (PSMA), also known as glutamate carboxypeptidase II or FOLH1. PSMA is a well-validated target that is significantly overexpressed in prostate cancer cells, particularly in metastatic castration-resistant prostate cancer (mCRPC). Upon administration, 212Pb-PS0001 binds to PSMA-expressing tumor cells and delivers high-linear-energy-transfer (LET) alpha radiation, which induces lethal double-strand DNA breaks and potent cytotoxicity with minimal damage to surrounding healthy tissue. In addition to its direct anti-tumor activity, 212Pb-PS0001 is being studied for its potential to modulate the tumor microenvironment and induce a systemic anti-tumor immune response.
