2155-14 is a preclinical small molecule inhibitor designed to target the spliceosomal proteins heterogeneous nuclear ribonucleoprotein H1 (hnRNPH1) and H2 (hnRNPH2). These proteins play a critical role in the alternative splicing of pre-mRNA and are frequently overexpressed in aggressive malignancies, including melanoma. By downregulating hnRNPH1/H2, 2155-14 disrupts the splicing machinery necessary for tumor cell survival and proliferation. Preclinical studies have demonstrated that 2155-14 significantly reduces tumor growth in both BRAF inhibitor-sensitive and BRAF/NRAS inhibitor-resistant melanoma xenograft models. Furthermore, the compound has shown an ability to enhance the antitumor immune response by increasing the infiltration of immune cells into the tumor microenvironment, suggesting potential for use as a monotherapy or in combination with existing immunotherapies.
