22-Rap is a preclinical-stage immunoRNase (antibody-toxin conjugate) developed by IBC Pharmaceuticals and Immunomedics (later acquired by Gilead Sciences). It is generated using the proprietary Dock-and-Lock (DNL) platform technology, which site-specifically tethers four copies of ranpirnase (Rap), an amphibian ribonuclease derived from Rana pipiens, to the humanized anti-CD22 monoclonal antibody epratuzumab (hLL2). 22-Rap is designed to target CD22-expressing B-cell malignancies, such as B-cell lymphomas and leukemias (including chronic lymphocytic leukemia and Burkitt lymphoma). Upon binding to CD22, the conjugate is rapidly internalized, delivering ranpirnase to the cytosol where it degrades tRNA, inhibits protein synthesis, and induces apoptosis in cancer cells.
