"222" is a combination analgesic product primarily marketed in Canada, consisting of acetylsalicylic acid (aspirin), caffeine, and codeine phosphate. Acetylsalicylic acid provides analgesic, antipyretic, and anti-inflammatory effects by irreversibly inhibiting cyclooxygenase enzymes (COX-1 and COX-2), which reduces the synthesis of pro-inflammatory prostaglandins. Codeine phosphate is an opioid analgesic that acts as a weak agonist at mu-opioid receptors, altering the central perception of pain. Caffeine is included as an adjuvant to enhance the analgesic efficacy of the other components. Historically developed by Charles E. Frosst & Co. (which became Merck Frosst), the brand was part of the Johnson & Johnson-Merck Consumer Pharmaceuticals joint venture until Johnson & Johnson acquired full ownership in 2011. It is indicated for the relief of mild to moderate pain and the reduction of fever.
