Overview

225Ac-A9-0642 is a therapeutic radiopharmaceutical in development by Alpha-9 Oncology for the treatment of breast and prostate cancers. The agent consists of a proprietary ligand, A9-0642, which specifically targets the Gastrin-Releasing Peptide Receptor (GRPR), a G protein-coupled receptor frequently overexpressed in various solid tumors. The ligand is conjugated to Actinium-225 (225Ac), a potent alpha-emitting radioisotope. Upon binding to GRPR on the surface of cancer cells, the complex is internalized, allowing the 225Ac to deliver high-linear energy transfer (LET) alpha particles that induce lethal double-stranded DNA breaks. This targeted approach aims to maximize tumor cell killing while sparing adjacent healthy tissues. The program is currently in the IND-enabling phase.
