225Ac-bisPSMA is a targeted alpha therapy (TAT) radiopharmaceutical in preclinical development by Clarity Pharmaceuticals for the treatment of prostate cancer. It utilizes Clarity's proprietary bisPSMA platform, which features a dual-targeting ligand designed to increase uptake and retention in tumors expressing Prostate-Specific Membrane Antigen (PSMA). The compound is labeled with actinium-225 (225Ac), a high-energy alpha-emitting radioisotope that delivers potent, localized radiation to cancer cells. Upon binding to PSMA-positive cells, the alpha particles induce irreparable double-stranded DNA breaks, leading to cell death while minimizing damage to surrounding healthy tissue. This agent is intended to complement Clarity's copper-based theranostic products by providing a therapeutic approach for patients with metastatic castration-resistant prostate cancer (mCRPC).
