Drug intelligence / Profile preview
225Ac-CLDN18.2 (also known as PD-29875T) is a novel, first-in-class peptide-radioisotope conjugate (radioligand therapy) being developed by PeptiDream and its subsidiary PDRadiopharma. It consists of a highly selective macrocyclic peptide targeting Claudin 18.2 (CLDN18.2) conjugated to the alpha-emitting radioisotope actinium-225 (225Ac). Discovered using PeptiDream's proprietary Peptide Discovery Platform System (PDPS) technology, the therapeutic is designed to selectively deliver high-energy alpha radiation to CLDN18.2-expressing tumor cells, inducing double-strand DNA breaks and cell death while sparing healthy tissues. 225Ac-CLDN18.2 is being developed alongside a paired diagnostic imaging agent, 64Cu-CLDN18.2 (64Cu-PD-29875D), as a theranostic pair for the treatment and diagnosis of gastric cancer and other CLDN18.2-positive solid tumors, such as pancreatic and biliary tract cancers. It is currently in preclinical and IND-enabling development.
