225Ac-DOTA-anti-PD-L1 antibody is a preclinical radioimmunotherapy (RIT) compound consisting of an anti-PD-L1 (Programmed Death-Ligand 1) monoclonal antibody conjugated to the macrocyclic chelator DOTA and labeled with the alpha-emitting radionuclide Actinium-225 (225Ac). Developed by researchers at Johns Hopkins University, this radioimmunoconjugate is designed to deliver targeted high linear energy transfer (high-LET) alpha-particle radiation directly to the tumor microenvironment by binding to PD-L1-expressing cells. It has been evaluated preclinically in immunocompetent murine models of metastatic breast cancer to assess its pharmacokinetics, biodistribution, and dosimetry, demonstrating potential for targeted alpha therapy (TAT) in PD-L1-expressing solid tumors.
