225Ac-FAP-VG-02 is a radionuclide drug conjugate (RDC) being developed by VitsGen Therapeutics for the treatment of tumors expressing fibroblast activation protein (FAP). The drug consists of a targeting moiety specific for FAP conjugated to the alpha-emitting radioisotope Actinium-225. FAP is highly expressed in the cancer-associated fibroblasts (CAFs) of the tumor microenvironment in various solid tumors. By targeting FAP, 225Ac-FAP-VG-02 delivers potent alpha-particle radiation directly to the tumor site, inducing double-strand DNA breaks and subsequent cell death in both FAP-positive cells and neighboring tumor cells via the bystander effect. It is currently in the preclinical stage of development.
