Drug intelligence / Profile preview
225Ac-NM600 is an investigational targeted alpha-particle therapy (TAT) composed of the alpha-emitting radioisotope actinium-225 conjugated to NM600, a small-molecule alkylphosphocholine (APC) analog. NM600 serves as a tumor-targeting vector by selectively binding to phospholipid ethers that are overexpressed in the plasma membranes of a wide variety of cancer cells and within the tumor microenvironment. Upon binding, the actinium-225 isotope releases high-energy alpha particles with a short path length (50-100 μm), inducing complex, lethal double-strand DNA breaks in the target tumor cells while minimizing collateral damage to adjacent healthy tissues. This approach is designed to treat both primary macroscopic tumors and disseminated microscopic metastatic disease. 225Ac-NM600 is being explored for the treatment of advanced solid tumors, particularly in combination with immune checkpoint inhibitors to overcome therapeutic resistance and promote a robust anti-tumor immune response.
