225Ac-NM600 + 90Y-NM600 is an experimental combination radiopharmaceutical therapy (RPT) designed to treat advanced and metastatic solid tumors. It consists of two distinct radiolabeled versions of NM600, a small-molecule alkylphosphocholine (APC) analog that selectively targets lipid rafts—specialized membrane microdomains that are overexpressed in a wide variety of cancer cells. By combining the alpha-emitter Actinium-225 (225Ac) with the beta-emitter Yttrium-90 (90Y), the therapy leverages the unique physical properties of both radionuclides: 225Ac provides high linear energy transfer (LET) radiation over a very short range, making it ideal for eliminating microscopic metastatic disease, while 90Y delivers lower LET radiation over a longer distance, which is more effective for debulking macroscopic primary tumors. This dual-radionuclide approach is often studied in conjunction with immune checkpoint inhibitors to enhance anti-tumor immune responses and overcome treatment resistance.
