225Ac-TR2205 is an experimental targeted alpha-therapy (TAT) radiopharmaceutical being developed for the treatment of advanced metastatic triple-negative breast cancer (TNBC). It consists of the alpha-emitting radioisotope Actinium-225 (225Ac) conjugated to TR2205, a monoclonal antibody that specifically targets Trophoblast cell-surface antigen 2 (TROP2), a protein frequently overexpressed in TNBC. The mechanism of action involves the antibody-mediated delivery of Actinium-225 to tumor cells, where the isotope undergoes decay, emitting high-energy alpha particles. These particles cause lethal double-strand DNA breaks within a short range, effectively killing the cancer cells while sparing distant healthy tissue. The drug is currently undergoing exploratory Phase 1 clinical trials sponsored by Xiaorong Sun to evaluate its safety, tolerability, and dosimetry.
