22ALA-CART is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat B-cell malignancies, including acute lymphoblastic leukemia (ALL). It utilizes the Adjunctive LAT Activating (ALA-CART) platform, which consists of a bicistronic construct expressing a second-generation CD22-BBz CAR alongside a novel signaling component incorporating the intracellular domain of the Linker for Activation of T cells (LAT). This design aims to overcome signaling deficits in standard CAR T cells, particularly when encountering low levels of antigen, by enhancing LAT activation. Preclinical studies indicate that 22ALA-CART induces global transcriptional changes that promote a less differentiated, stem-cell memory phenotype, leading to improved potency and persistence compared to standard CD22-directed CAR T cells.
