22B1 is a mouse monoclonal antibody that acts as a stimulatory agonist of the thyrotropin receptor (TSHR). It was developed as a research tool to characterize the molecular basis of the pathological antibody response in Graves' disease. 22B1 binds to the TSHR and activates both canonical and non-canonical NF-κB signaling pathways, leading to the induction of chemokines such as CCL2, CCL19, and CCL5 in human thyrocytes. This activation mimics the effect of stimulatory autoantibodies found in Graves' disease patients, which drive immune imbalance and thyroid hyperplasia. In preclinical studies, 22B1 and related antibodies have demonstrated the ability to induce elevated serum thyroxine and thyroid follicular epithelial hyperplasia, effectively modeling the clinical presentation of hyperthyroidism.
