22C7 is a high-affinity neutralizing monoclonal antibody targeting interleukin-25 (IL-25), also known as IL-17E. IL-25 is a type 2 cytokine and a member of the IL-17 family that initiates innate and adaptive type 2 immune responses by signaling through the IL-17RA-IL-17RB receptor complex. Developed by researchers at Pfizer, 22C7 was originally generated via mouse immunization and subsequently humanized using a complementarity determining region (CDR)-grafting approach onto human germline frameworks (specifically the VH3-23/Vk1-39 graft). In preclinical studies, 22C7 demonstrated potent neutralization of both human and mouse IL-25, effectively reducing cellular recruitment in air pouch models and ameliorating pathology in Aldara-induced psoriasiform skin inflammation. It is being investigated for its therapeutic potential in a spectrum of IL-25-mediated diseases, including asthma, atopic dermatitis, and various autoimmune conditions.
