Overview

22E5 is a monoclonal antibody that specifically targets murine thymic stromal lymphopoietin (TSLP). It serves as a mouse-specific surrogate or tool compound for solrikitug (formerly UPB-101), an optimized human-specific anti-TSLP monoclonal antibody. TSLP is an epithelial-derived cytokine, or alarmin, that acts as an upstream initiator of the allergic inflammatory cascade by activating dendritic cells and mast cells, thereby promoting Type 2 inflammation. In preclinical studies using ovalbumin (OVA)-induced mouse models of asthma, 22E5 has demonstrated the ability to significantly reduce airway hyperresponsiveness (AHR), bronchoalveolar lavage fluid (BALF) eosinophils, and TARC (CCL17) levels. It is utilized by Upstream Bio to evaluate the attenuation of key inflammatory markers in murine models, providing proof-of-concept data that supports the differentiated efficacy of solrikitug for the treatment of asthma and other eosinophilic diseases.
