264RAD is a human monoclonal antibody developed by AstraZeneca that targets the integrin alpha-v beta-6 (αvβ6), with additional inhibitory activity against integrin alpha-v beta-8 (αvβ8). These integrins are epithelial-specific receptors that are upregulated during tissue remodeling and in various cancers, where they promote tumor cell growth, migration, and invasion, and facilitate the activation of TGF-β. By blocking these pathways, 264RAD inhibits tumor progression and modulates the tumor microenvironment, particularly by reducing desmoplasia and suppressing pro-tumorigenic signaling. Preclinical studies have demonstrated its efficacy in models of pancreatic ductal adenocarcinoma (PDAC), breast cancer, and oral cancer, especially when used in combination with chemotherapy agents like gemcitabine.
