Overview

2A3-CAR T-cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target members of the carcinoembryonic antigen-related cell adhesion molecule (CEACAM) family. Developed by 4Cell Therapies, the construct utilizes a camelid single-domain antibody (sdAb), or nanobody, derived from the 2A3 clone. While primarily targeting CEACAM6, the 2A3-CAR exhibits cross-reactivity with CEACAM5, allowing for dual-targeting of these antigens which are frequently overexpressed in solid tumors such as pancreatic ductal adenocarcinoma and breast cancer. The CAR architecture incorporates CD28 and CD3ζ signaling domains to drive T-cell activation and cytotoxicity upon antigen engagement. Preclinical data indicate significant antitumor activity in both in vitro assays and in vivo xenograft models.
