2A5 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD70, a member of the tumor necrosis factor (TNF) superfamily that is overexpressed in various malignancies, including renal cell carcinoma (RCC). The 2A5 construct utilizes a camelid-derived single-domain antibody (VHH or nanobody) as the antigen-binding domain, which offers potential advantages over traditional single-chain variable fragments (scFv), such as smaller size, increased stability, and potentially lower immunogenicity. Preclinical studies have demonstrated that 2A5 CAR-T cells exhibit potent cytotoxicity against CD70-positive tumor cells, high cytokine production (IFN-γ, IL-2, and TNF-α), and superior in vivo expansion and persistence compared to scFv-based CAR-T cells. This therapy is currently being investigated for its potential to treat RCC and other CD70-expressing solid and hematological tumors.
