2A5 VHH CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD70, a member of the tumor necrosis factor (TNF) superfamily that is overexpressed in various cancers, including renal cell carcinoma (RCC), while showing limited expression in healthy tissues. Developed by HRAIN Biotechnology, the therapy incorporates a VHH (variable domain of heavy-chain-only antibody) binder, also known as a nanobody, which is derived from an immunized alpaca library. In preclinical models, 2A5 VHH CAR-T cells demonstrated potent in vitro cytotoxicity and in vivo anti-tumor activity, including significant expansion and cytokine production (IL-2) upon antigen encounter. The use of VHH binders in CAR-T construction is intended to provide high affinity and stability while potentially reducing the risk of immunogenicity compared to traditional scFv-based CARs.
